A case of severe glutathione synthetase deficiency with novel GSS mutations BJMBR
Xia,H.; Ye,J.; Wang,L.; Zhu,J.; He,Z..
Glutathione synthetase deficiency (GSSD) is a rare inborn error of glutathione metabolism with autosomal recessive inheritance. The severe form of the disease is characterized by acute metabolic acidosis, usually present in the neonatal period with hemolytic anemia and progressive encephalopathy. A case of a male newborn infant who had severe metabolic acidosis with high anion gap, hemolytic anemia, and hyperbilirubinemia is reported. A high level of 5-oxoproline was detected in his urine and a diagnosis of generalized GSSD was made. DNA sequence analysis revealed the infant to be compound heterozygous with two mutations, c.738dupG in exon 8 of GSS gene resulting in p.S247fs and a repetitive sequence in exon 3 of GSS gene. Treatment after diagnosis of GSSD...

A model on biological flow of phosphorus in growing pigs Arq. Bras. Med. Vet. Zootec.
Lopes,J.B.; Moreira,J.A.; Kebreab,E.; Vitti,D.M.S.S.; Abdalla,A.L.; Crompton,L.A.; France,J..
Ten crossbred barrows weighting 20kg were allocated in five groups fed and on diets consisted of different total phosphorus (P) levels: 0.30%, 0.40%, 0.51%, 0.65%, and 0.73%. The animals were intravenously injected 7.4MBq of radioactive P (32P) and after seven days they were slaughtered for further analysis. The evaluated parameters were intake, excretion, specific activities, and flow of P in compartments (1-gut, 2-blood, 3-bone, and 4-soft tissue). Besides fecal losses, the kidneys played an import role in P regulation. The flows of P from tissue and gut to the blood were affected by P intake.

Acephate and methamidophos residues in greenhouse and in field grown tomatoes Horticultura Brasileira
Trevizan,Luiz Roberto P.; Baptista,Gilberto C. de; Papa,Geraldo.
Increasing horticultural production under protected cultivation requires more detailed studies about the occurence of pesticide residues, due to the special and peculiar environment under controlled conditions, in which some pests, that are of little concern in the field, become important. The insecticide acephate and its methamidophos metabolite residues were evaluated in greenhouse-grown tomatoes and compared to an open-field tomato crop. The treatments: (a) check; (b) one application of 75 g a.i. acephate.100 L-1 water; (c) one application of 150 g a.i.100 L-1 water; (d) four applications of 75 g a.i.100 L-1 water were evaluated. Fruit, leaf and soil samples were taken at (-1), zero, 1, 3, 7, 14, 21 days after the last or only application. The...

Activity and composition of the benthic fauna in the Whittard Canyon and the adjacent continental slope (NE Atlantic) ArchiMer
Duineveld, Gérard; Lavaleye, Marc; Berghuis, Eilke; De Wilde, Peter.
We compared the sediment and its community on the Celtic continental slope (Goban Spur) with those in a branch of the nearby Whittard Canyon in search for evidence of canyon mediated transport of (labile) organic matter. We studied the megabenthos and macrobenthos biomass and taxonomic composition, measured in situ sediment community oxygen consumption and determined sediment concentrations of particulate organic carbon, phytopigments, and nucleic acids. While the sediment community and activity on the canyon fan was similar to that on the abyssal station near Goban Spur, the sediment within the canyon had relatively enhanced sediment community oxygen consumption rates and higher levels of phytopigments. particulate organic carbon and nucleic acids,...

Advances in improving mammalian cells metabolism for recombinant protein production Electron. J. Biotechnol.
Altamirano,Claudia; Berrios,Julio; Vergara,Mauricio; Becerra,Silvana.
Background: The production of recombinant proteins for therapeutic use represents a great impact on the biotechnology industry. In this context, established mammalian cell lines, especially CHO cells, have become a standard system for the production of such proteins. Their ability to properly configure and excrete proteins in functional form is an enormous advantage which should be contrasted with their inherent technological limitations. These cell systems exhibit a metabolic behaviour associated with elevated cell proliferation which involves a high consumption of glucose and glutamine, resulting in the rapid depletion of these nutrients in the medium and the accumulation of ammonium and lactate. Both phenomena contribute to the limitation of cell...

Allelochemicals from Alexandrium minutum induce rapid inhibition of metabolism and modify the membranes from Chaetoceros muelleri ArchiMer
Long, Marc; Tallec, Kevin; Soudant, Philippe; Le Grand, Fabienne; Donval, Anne; Lambert, Christophe; Sarthou, Geraldine; Jolley, Dianne F.; Hegaret, Helene.
Allelochemical interactions are likely to be a contributing factor explaining the success of large blooms of the harmful marine dinoflagellate Alexandrium, however, the physiological mechanisms of allelochemical interactions remain poorly described. Here we investigated the sub-lethal effects (on an hourly scale) of a filtrate containing allelochemicals from Alexandrium minutum on the physiology of the common diatom Chaetoceros muelleri. The filtrate induced deleterious effects to the diatom physiology within only 30 min of exposure. Esterase activity and photosynthesis were drastically inhibited, with up to 34% of the population being metabolically inactive and up to 30% reduction in photosystem II quantum yield when exposed to the filtrate. In addition,...

Allocation du carbone et métabolisme azoté chez l'haptophyte Tisochrysis lutea ArchiMer
Garnier, Matthieu.
The aim of this thesis is to improve knowledge on mechanisms involved in the response to nitrogen limitation and in lipid accumulation in the microalgae haptophyte Tisochrysis lutea. The wild type strain and a lipid accumulating mutant strain were grown under different nitrogen limitation and starvation and analyzed by functional genomics. Four genes of high-affinity nitrate/nitrite transporter (Nrt2) were identified and characterized to reveal the mechanisms involved in mineral absorption in this species. Transcriptomes of both strains were sequenced and proteins affected by nitrogen starvation and differentially expressed between the two strains were identified. We so identified the functions regulated by nitrogen deficiency and potentially involved in...

Allosteric regulation of 6-phosphofructo-1-kinase activity of fat body and flight muscle from the bloodsucking bug Rhodnius prolixus Anais da ABC (AABC)
Alves,Gutemberg G.; Marinho-Carvalho,Monica M.; Atella,Georgia C.; Silva-Neto,Mario A.C.; Sola-Penna,Mauro.
6-phosphofructo-1-kinase (phosphofructokinase; PFK) activity from Rhodnius prolixus, a haematophagous insect which is usually a poor flyer, was measured and compared in two metabolically active tissues - flight muscle and fat body. The activity of this important regulatory glycolytic enzyme was much more pronounced in muscle (15.1 ± 1.4 U/mg) than in fat body extracts (3.6±0.4 U/mg), although the latter presented higher levels of enzyme per protein content, as measured by western-blotting. Muscle extracts are more responsible than fat body to ATP and fructose 6-phosphate, both substrates of PFK. Allosteric regulation exerted by different effectors such as ADP, AMP and fructose 2,6-phosphate presented a singular pattern for each tissue. Optimal pH (8.0-8.5)...

Antioxidants and dairy production: the example of flax R. Bras. Zootec.
Petit,Hélène V..
This manuscript reports on the main problems decreasing productivity of dairy cows (e.g. fatty liver syndrome and poor fertility) and how antioxidants could enhance it. High producing dairy cows are prone to oxidative stress, and the situation can be exacerbated under certain environmental, physiological, and dietary conditions. Antioxidants have important effects on the expression of genes involved in the antioxidant status, which may enhance animal health and reproduction. Moreover, antioxidants may contribute to decrease the incidence of spontaneous oxidized flavour in milk enriched in polyunsaturated fatty acids. Plant lignans are strong antioxidants and flax is the richest source of plant lignans. Flax lignans are converted in the mammalian lignans...

Application of a micro-respirometric volumetric method to respiratory measurements of larvae of the Pacific oyster Crassostrea gigas ArchiMer
Goulletquer, Philippe; Wolowicz, Maciej; Latala, Adam; Brown, Craig; Cragg, Simon.
The application of a simple, fast and reliable volumetric microrespirometric method to assess respiration of bivalve larvae is discussed. As a model, C. gigas larvae of various sizes were used. Metabolic activity of veliger larvae was assessed by measuring respiratory rate for use in ecophysiological modelling. As an example of the application of this approach, additional measurements of veliger respiratory rates were carried out to assess the effect on larval metabolism of different concentrations of leachate from wood treated with chromated copper-arsenate (CCA). Veligers of Crassostrea gigas (length from 95 to 331 pm) were fed with a mixture of Isochrysis galbana) and Chaetoceros pumilum cells. Experiments were performed in a 20 degreesC constant...

Associations between tissue fatty acid composition and physiological traits of performance and metabolism in the seabass (Dicentrarchus labrax) ArchiMer
Chatelier, Aurelien; Mckenzie, David; Prinet, Aurelie; Galois, Robert; Robin, Jean; Zambonino, Jose-luis; Claireaux, Guy.
Seabass were fed for 4 months with diets where the lipid was provided as either canola oil (CO), palm oil ( PO) or fish oil (FO), to generate diversity in their tissue fatty acid (FA) composition and investigate how this influenced major traits of exercise performance, cardiac performance and respiratory metabolism. In particular, based upon previous observations, we investigated the hypothesis that enriching the fish tissues with oleic and linoleic acids (OA, 18:1n-9 and LA, 18:2n-6, respectively) from the CO and PO diets would improve maximum exercise and cardiac performance, and increase aerobic metabolic scope. This proved to be the case; exercise respirometry on bass fitted with cardiac flow probes revealed that those fed CO and PO diets had a...

Bioenergética del besugo Pagrus pagrus. OceanDocs
Aristizabal, E.O..
The utilization of the energy of food by red porgy (Pagrus pagrus Linnaeus, 1758) from the Argentine Sea (SW Atlantic) was studied. From the energetic point of view, it is proposed that the strategy of patitioning, utilization and storage of the energy of food, is an indicator of the organism response to the environmental conditions. The main objective was to know the proportion of the energy of food derived to the different components of the bioenergetics budget. In order to estimate the monthly energy investment in reproductive growth, chemical analysis of gonads from fishes caught by the fishery fleet and from laboratory studies were performed. Two indexes were used to calculate the energy investment in female ovaries: the proportion of net daily energy...

Biomass distribution and physiological capabilities of bacteria in the water column above a sea grass system. ArchiMer
Velimirov, B; Herndl, G; Kavka, G.
The distribution pattern of bacteria in the water column above and within a Posidonia oceanica bed near Ischia (off the Gulf of Naples) was investigated and correlated with DOC and POM data for autumn and winter. Data from samples of 5 stations along a transect from shallow to deep showed rather low bacterial densities (10 super(4).ml super(-1)) for both seasons. The number of attached bacteria was higher in late winter as compared to autumn when the leaves are shed from the stands. Except for occasional peaks DOC ranged between 1.5 and 4.5 mg C/1 while POM concentrations ranged form 4 to 14 mg AFDW.1 super(-1). Hydrolytic capabilities of isolated bacterial colonies showed that more carbohydrate substrates could be fermented in winter as compared to...

Biomass, metabolism and nutrition of zooplankton in the Gulf of Lions (NW Mediterranean) ArchiMer
Gaudy, R; Youssara, F; Diaz, F; Raimbault, P.
Zooplankton biomass was measured at 20 stations located along five coastal-offshore transects in the Gulf of Lions (NW Mediterranean) during spring 1998 and winter 1999, using two nets equipped with 200 and 80 Pm mesh. Biomass was twice as abundant in spring. It increased toward the coast and the west of the gulf and was positively correlated to chlorophyll, primary production and organic particulate matter and negatively to temperature and salinity. The enrichment effect of Rhone river input and the oligotrophic influence of the north Mediterranean current (NMC) were the main distribution factors of zooplankton biomass. Higher biomass was obtained in coastal stations with the 80 mum net, which sampled more efficiently young larval stages and nauplii....
